Question

Is there a taqueria anywhere that is genuinely worth flying for?

Serious question. I have a week off in November and no plan. I am willing to build the whole trip around one place if that place is actually worth it. Not looking for the famous ones that turn up in every list. Looking for the one you would personally get on a plane for.

Yes, and it is not a taqueria, it is a market stall in Oaxaca that does one thing with one chile and closes when it runs out. The honest answer to your question though is that you should not fly for a single place, because the good version of this trip is eating twelve mediocre things and two extraordinary ones in the same neighbourhood. Pick a city with depth rather than a restaurant with a reputation. Oaxaca or Puebla, both have more than a week in them.

Agreed on Oaxaca, and go for the mezcalerias as much as the food. Half the good eating I did there was because someone in a bar told me where to go.

Puebla for mole and I will not be taking questions. Two days minimum, and eat at least four different ones so you can tell what the differences actually are.
